Product Overview

When biologically active proteins are required for applications such as Co-IP, pull-down assays, or enzymatic activity analysis, conventional strongly denaturing lysis methods disrupt protein structure and intermolecular interactions. In contrast, the extraction of functional proteins under non-denaturing conditions involves several major technical challenges:

1. Low Yield of Membrane Proteins

Mild nonionic detergents alone are insufficient to efficiently solubilize hydrophobic membrane proteins, resulting in substantial loss of membrane-associated species during native extraction.

2. Interference from Genomic DNA and Lipids

Upon tissue disruption, released genomic DNA and lipids cause substantial sample viscosity and reduced clarity in gentle non-denaturing buffers, which severely compromise the efficiency and reliability of downstream immunoprecipitation and related assays.

3. Instability of Protein Activity

Extended extraction procedures may fail to adequately suppress protease activity, leading to degradation and loss of native function.
