Product Overview

Routine whole cell or nuclear lysis is often insufficient for histone extraction, as histones are tightly bound to DNA and easily lose post-translational modification information. The main challenges of histone extraction include:

1. Strong DNA-Protein Interactions

Robust DNA-protein interactions can trap histones in highly viscous chromatin fragments, making separation difficult and reducing the amount of soluble histone available for antibody based or MS based assays.

2. Labile Post Translational Modifications

Histone acetylation, methylation, phosphorylation, and other PTMs are sensitive to deacetylases, phosphatases, and proteases activated during lysis, which can alter modification patterns and compromise epigenetic readouts.

3. High DNA Content and Sample Viscosity

Large amounts of genomic DNA released during chromatin disruption increase viscosity, complicate pipetting and sample handling, and can interfere with gel separation and LC-MS performance.

4. Contamination by Non Histone Nuclear Proteins

Conventional nuclear extraction often co isolates abundant non histone proteins, reducing histone purity and increasing background in Western blotting, ChIP related assays, and mass spectrometry.
