MtoZ Biolabs' Human Immunoglobulin A (IgA) ELISA Kit is a high-specificity immunoassay kit designed for the quantitative detection of human IgA. It accurately measures IgA levels in serum, plasma, saliva, and other biological samples. This kit is widely used in mucosal immunity research, autoimmune disorder investigation, IgA nephropathy studies, and vaccine immunogenicity evaluation.

Product Overview

IgA is the predominant antibody class at mucosal surfaces and the second most abundant immunoglobulin in serum. Accurate quantification is essential for immune function analysis, yet presents distinct challenges:

1. Multiple Structural Forms

IgA exists as monomeric (serum) and dimeric secretory forms (sIgA), each differing in molecular weight and epitope accessibility, complicating standardized detection.

2. Subclass Complexity

IgA1 and IgA2 differ in hinge region structure and glycosylation patterns, affecting antibody binding efficiency and assay specificity.

3. Cross-Reactivity Risk

Structural homology with other immunoglobulin isotypes increases the risk of non-specific binding, particularly in samples with elevated IgG or IgM concentrations.

4. Complex Mucosal Matrices

Saliva, bronchoalveolar lavage fluid, and intestinal secretions contain proteases, mucins, and high-viscosity components that can degrade IgA and interfere with assay performance.

5. Wide Dynamic Range Requirement

IgA concentrations vary significantly across sample types and disease states, requiring broad linear range coverage to avoid dilution errors and Hook Effect.

Advantages

 Sensitivity

LOD reaches the ng/mL level, enabling accurate quantification of IgA in both serum and low-concentration mucosal samples such as saliva and secretory fluids.

 Specificity

Antibodies target epitopes unique to the IgA heavy chain (α chain), with minimal cross-reactivity to IgG, IgM, IgE, and IgD, ensuring accurate detection in complex matrices containing multiple immunoglobulin isotypes.

Precision

Intra-assay (CV%<8%) and inter-assay (CV%<10%) CVs are within acceptable limits.
