During protein extraction, both protein integrity and post-translational modification status are highly susceptible to interference from endogenous enzymatic activities, mainly including the following challenges:

1. Rapid Activation of Endogenous Proteases

Upon cell lysis, multiple proteases are instantly released and activated, leading to non-specific degradation of target proteins and compromising molecular weight determination and quantitative accuracy.

 

2. Loss of Modification States Caused by Phosphatases

Endogenous phosphatases can rapidly remove phosphorylation modifications, resulting in distorted signaling information and interference with downstream functional analyses.
