MtoZ Biolabs' Phosphatase Inhibitor is composed of multiple active components targeting both Ser/Thr and Tyr phosphatases. Product Overview

Maintaining native protein phosphorylation levels becomes particularly challenging during in vitro sample processing, where non-specific dephosphorylation readily occurs. The main challenges include:

1. Rapid Loss of Phosphorylation

Upon cell lysis, multiple Ser/Thr and Tyr phosphatases are instantly activated, causing key phosphorylation sites to disappear within minutes and compromising accurate signaling assessments.

2. High Abundance of Endogenous Phosphatases

Crude lysates contain numerous potent phosphatases capable of altering protein modification states, resulting in deviations from true cellular phosphorylation levels.

3. Increased Instability During Handling

Temperature fluctuations, mixing, or prolonged lysis steps accelerate dephosphorylation and reduce reproducibility across experiments.

4. Distortion of Downstream Analyses

Loss of phosphorylation leads to weakened Western blot signals, unreliable phospho-antibody detection, and significant bias in phosphoproteomic identification and quantification.

5. Difficulty Preserving Native Regulatory States

Without effective inhibition, samples cannot maintain their instantaneous cellular phosphorylation levels, hindering accurate evaluation of kinase activity and signaling dynamics.

Advantages

✅ High Efficiency

A composite Ser/Thr and Tyr phosphatase inhibitor system rapidly blocks endogenous phosphatase activity and stabilizes phosphorylation sites.

Preservation

Effectively prevents non-specific dephosphorylation during lysis and handling, maintaining phosphorylation levels closer to in-cell conditions.

✅ Compatibility

Suitable for Western blotting, IF, IP/Co-IP, and phosphopeptide mass spectrometry, ensuring reliable qualitative and quantitative outcomes.
