Sperm chromatin is extremely compact, with most histones replaced by protamines. This unique structural organization presents significant challenges for histone extraction:
1. Ultracondensed Chromatin Structure
Sperm nuclear proteins are dominated by protamines, leaving histones at very low abundance and tightly embedded. Conventional lysis methods cannot sufficiently disrupt the structure or release target histones.
2. Highly Resilient Membrane and Acrosomal Structures
The sperm plasma membrane and acrosome are exceptionally robust, and standard physical or chemical lysis is often insufficient to fully disrupt the cell, resulting in low extraction efficiency.
