1. Lysis buffer 1 should be freshly prepared before use by mixing Lysis buffer A and B components.

Protocol

The entire workflow can be performed under standard laboratory conditions without the need for specialized equipment. The following are the recommended steps:

1. Sample Preparation

Pipette 100-200 μL of serum sample into a clean EP tube (dilution may be applied to optimize precipitation efficiency).

  

2. High-Abundance Protein Removal

(1) Take an appropriate amount of pre-washed magnetic beads and add them to the corresponding volume of serum sample.

(2) Mix thoroughly and incubate on a rotator for 10-15 minutes.

(3) Use a magnetic stand to separate the beads and wash them.

(4) Elute the low-abundance proteins from the beads.

  

3. Digestion of Low-Abundance Proteins

(1) Dilute the protein solution with digestion buffer and perform overnight digestion.

(2) After digestion, collect the peptide solution using the provided elution buffer.

  

4. Digestion Termination and Sample Cleanup (Optional)
