Micro-scale protein samples typically contain low protein amounts and are more likely to carry residual salts and buffer components, resulting in a higher proportion of background constituents. These characteristics pose significant challenges to traditional LysN digestion and desalting workflows:

 

1. Micro-Scale Recovery Loss

After extraction, the overall amount of micro-scale samples is limited. In traditional stepwise workflows, repeated tube changes, transfers, and multiple opening steps more readily cause adsorption, residual retention, and sample loss, making low-abundance components more difficult to preserve.
