The C-Trap LysN Digestion and Desalting Kit launched by MtoZ Biolabs is designed for complex protein samples, enabling LysN-directed digestion integrated with desalting and helping to improve the efficiency of N-terminus-related proteomics analysis.

Product Overview

During proteomics mass spectrometry sample preparation, samples originate from diverse sources and are often accompanied by residual salts, detergents, and denaturants. These characteristics introduce challenges to the stability and consistency of LysN digestion and desalting workflows:

1. Low Input Variability

In traditional stepwise workflows, microscale samples require repeated opening and transfer, which more readily leads to adsorption, residual retention, and irreversible loss, affecting the stable detection of N-terminus-related peptides.

2. Differences in Site Accessibility

In proteins with compact conformations, aggregation, or extensive modifications, lysine site accessibility varies, making localized missed cleavages more likely under conventional conditions and resulting in shifts in peptide yield and distribution.

3. Matrix Heterogeneity

Inconsistent levels of residual salts, detergents, and denaturants among samples can affect LysN digestion kinetics and increase missed cleavages, leading to reduced comparability of batch results.

4. Unstable Desalting Recovery

Residual salts and interfering components in extracts elevate background signals and cause ion suppression, while slight mismatches in traditional desalting conditions can also lead to fluctuations in peptide recovery, affecting data stability.

Advantages

✅ LysN-Directed Digestion

The system is optimized around the characteristic of LysN cleaving at the N-terminus of lysine residues, helping to achieve more consistent cleavage output and improving the stability of N-terminus-related information interpretation.

✅ Workflow Integration

The MtoZ Biolabs' C-Trap LysN Digestion and Desalting Kit integrates digestion and desalting within the same tube, reducing handling and lowering the risk of sample loss and contamination.

✅ Result Consistency

Standardized reaction and washing workflows reduce sources of operator-dependent variation, making them more suitable for the comparability and cross-batch stability requirements of batch samples.

✅ Integrated Desalting

The built-in desalting step simultaneously attenuates the impact of salts and interfering components, reducing ion suppression and background noise and allowing LC-MS/MS analysis to more readily reach a stable detection state.
