Product Overview

During the research, development, and production of biological products, BHK (Baby Hamster Kidney) cells are commonly used as host cells or expression systems. Effectively monitoring and removing their residual DNA is crucial for ensuring drug safety. However, this detection process faces several challenges:

1. DNA Fragmentation

Host genomic DNA often breaks into small fragments during processes like cell lysis or purification. Traditional detection methods may miss these small fragments, leading to underestimated values or inaccurate residual DNA levels.

2. Matrix Effects and PCR Inhibition

Samples like recombinant proteins, antibodies, and vaccines contain complex components (proteins, salts, buffers) that can inhibit PCR amplification, causing false negatives or inaccurate quantification.

3. Sensitivity for Trace Residues

After purification, residual DNA in the final product is extremely low (pg or fg levels). Detecting these small amounts requires highly sensitive methods with a good signal-to-noise ratio.

4. Fragment Distribution Risk Assessment

Regulatory agencies are focusing not only on the total amount of DNA but also its fragment size distribution. Longer fragments (>200 bp) carry a higher risk, making the detection of various fragment sizes challenging.

5. Interference from Species Similarity

BHK and CHO cells have similar genomic sequences, which can cause cross-reactivity if the detection targets are not specific enough, reducing assay accuracy.

Advantages

 Comprehensive Fragment Size Analysis

MtoZ Biolabs' BHK Residual DNA Fragment Detection Kit quantifies four fragment sizes (81bp, 134bp, 216bp and 589bp) in parallel.

High Sensitivity

Detection sensitivity down to the fg level across a broad dynamic range.

Integrated Internal Process Control (IPC)

Enables real-time monitoring of amplification performance and sample inhibition, ensuring data reliability.

High Analytical Specificity

BHK-specific primers and fluorescent probes provide excellent specificity with minimal cross-reactivity.

 Seamless Integration

Compatible with mainstream real-time PCR platforms and designed for use with the Host Cell Residual DNA Sample Preprocessing Kit, streamlining workflow integration.
