These characteristics pose significant challenges for Lys-C digestion and desalting:

1. Insufficient Digestion Stability

Under traditional conditions, it is difficult to uniformly control different salt concentrations, urea levels, and detergent backgrounds, which readily leads to fluctuations in digestion rate and missed cleavages, resulting in inconsistent peptide yields.
