The PNGase F Deglycosylation Kit from MtoZ Biolabs is specifically engineered for mass spectrometry and glycan profiling applications, providing an efficient and comprehensive solution for the complete removal of N-linked glycans from glycoproteins.

Product Overview

In glycoprotein structure and functional studies, biomarker discovery, and biopharmaceutical glycosylation analysis, the complete and specific removal of N-linked glycans is a critical sample preparation step for accurate mass spectrometric identification, glycan profiling, and downstream functional characterization. However, traditional methods or non-specialized enzymatic formulations often encounter several limitations, including:

1. Interference Introduced by Impure Enzyme Preparations

Conventional PNGase F formulations may contain contaminants such as Endo F1, Endo F2, or Endo F3, which can induce non-specific cleavage or partial hydrolysis of glycans. These reactions generate heterogeneous glycopeptide fragments that significantly compromise mass spectrometric data interpretation and glycan assignment accuracy.

2. Restricted Reaction Conditions

Many commercial formulations function only under denaturing conditions and are unable to remove glycans while maintaining native protein conformations. This limitation hinders the study of native glycoprotein function, protein-protein interactions, and structural analyses.

3. Incomplete Deglycosylation

Suboptimal enzymatic activity or poorly defined reaction systems may result in incomplete removal of glycan moieties at glycosylation sites, causing missing peptide signals, quantitative bias, reduced proteomic coverage, and diminished reliability of analytical conclusions.

Advantages

✅ Ultra-High Purity Recombinant Enzyme

The PNGase F Deglycosylation Kit from MtoZ Biolabs provides recombinant PNGase F with ≥95% purity and is rigorously verified to be free of Endo F1, Endo F2, and Endo F3 enzymatic activities. This prevents off-target glycan cleavage and ensures high reaction specificity and accurate analytical results.

✅ Complete N-Glycan Removal Across All Glycan Types

The enzyme efficiently hydrolyzes all major classes of N-linked glycans, including high-mannose, hybrid, and complex types, by cleaving between the core GlcNAc residue and asparagine. Complete release of intact oligosaccharides enables downstream glycan profiling and mass spectrometry-based structural identification.

✅ Broad Applicability and Flexible Reaction Conditions

The kit supports both native and denaturing reaction environments. Users may select mild (native-state preserving) or strongly denaturing conditions depending on experimental requirements, expanding compatibility across functional and structural glycoprotein studies.

✅ Ready-to-Use Complete Formulation

Each kit includes high-concentration PNGase F enzyme (30,000 units) and a dedicated 10× reaction buffer, providing all essential components for a robust and reproducible deglycosylation workflow.
