Exosomes play critical roles in intercellular communication by transporting proteins, nucleic acids, lipids, and signaling molecules. Their inherent biocompatibility, stability, and ability to cross biological barriers make them highly attractive as both disease biomarkers and drug delivery vehicles. However, realizing the full therapeutic and diagnostic potential of exosomes relies on a thorough understanding of their in vivo distribution, cellular uptake, and organ-specific accumulation. To elucidate how exosomes behave in biological systems, precise labeling and tracking strategies are essential. These labeling and tracking approaches enable researchers to monitor exosome transport pathways, assess targeting specificity, and evaluate in vivo and in vitro functional outcomes.
Service at MtoZ Biolabs
MtoZ Biolabs offers a specialized Exosomes Labeling and Tracking Service that helps researchers visualize and quantify exosome dynamics with high sensitivity and accuracy. By integrating advanced labeling techniques with state-of-the-art imaging and analytical platforms, including fluorescence microscopes, electron microscopes, and flow cytometers, we provide tailored solutions for both in vivo and in vitro exosome studies. Our service enables comprehensive tracking of exosome biogenesis, release from parental cells, uptake by recipient cells, distribution in tissues, and levels in biological fluids.
We offer multiple labeling and tracking strategies optimized for different experimental settings:
💠Fluorescent Labeling: Ideal for both in vitro and in vivo applications. We utilize lipophilic dyes (e.g., PKH26, DiI, DiR), fluorescent nanomaterials, and genetically encoded fluorescent proteins, as well as antibody-based immunofluorescent labeling to visualize exosomes under imaging platforms.
💠Bioluminescence Imaging (BLI): Suitable for both in vivo and ex vivo tracking, this method uses luciferase-tagged exosomes to enable sensitive, real-time, and non-invasive visualization of exosome distribution and tissue accumulation.
💠Magnetic Resonance Imaging (MRI): Designed for in vivo imaging, we label exosomes with superparamagnetic iron oxide nanoparticles (SPIONs) to allow precise anatomical localization through high-resolution MRI.
💠Computed Tomography (CT): Applicable for in vivo use, CT-compatible contrast agents such as gold nanoparticles or iodine-labeled probes are employed to track exosomes in deep tissues with structural clarity.

Analysis Workflow
1. Sample Preparation and Exosome Isolation
Biological fluids or cell culture supernatants are processed using ultracentrifugation, size exclusion chromatography (SEC), or immunoaffinity-based methods to isolate pure exosomes.
2. Labeling Strategy Selection
Based on the research objective (in vivo vs. in vitro), appropriate labeling techniques are selected—such as fluorescent dyes, luciferase fusion proteins, immunogenic reporters, or fluorescent nanoparticles.
3. Exosome Labeling
Labeling is performed through direct surface tagging or parent cell engineering. Care is taken to preserve exosome integrity and function.
4. Tracking and Imaging
Labeled exosomes are administered to the desired biological system (cell culture or animal model), followed by real-time tracking using fluorescence microscopy, BLI, MRI, or CT.
5. Data Acquisition and Analysis
Quantitative and spatial data on exosome localization, uptake, and biodistribution are collected and analyzed using advanced imaging software and statistical tools.
