Membrane Proteomics Services

Composed primarily of lipid bilayers and embedded proteins, membranes are essential for maintaining homeostasis, mediating signal transduction, and supporting energy conversion in both prokaryotic and eukaryotic systems. Among membrane components, membrane proteins play especially critical roles. They serve as receptors, channels, transporters, enzymes, and adhesion molecules, participating in a wide range of physiological processes such as signal reception, nutrient transport, intercellular communication, and immune recognition. Despite representing approximately 30% of the proteome and being the target of over half of all approved drugs, membrane proteins remain one of the most technically challenging classes of proteins to study due to their hydrophobicity, low abundance, and structural complexity.

    Membrane proteomics focuses on the large-scale identification, quantification, and characterization of membrane-associated proteins. This field has become increasingly important for understanding disease mechanisms, identifying biomarkers, and discovering novel drug targets. Through high-resolution mass spectrometry and refined enrichment techniques, membrane proteomics enables researchers to elucidate dynamic changes in membrane composition under different physiological and pathological conditions.

    2. Membrane Protein Extraction, Separation, and Identification

    • Membrane Protein Extraction: We use optimized protocols involving organic solvent solubilization with detergents to effectively extract both integral and peripheral membrane proteins while preserving protein structure and yield.
    • Membrane Protein Separation and Identification: Extracted proteins or peptides are separated by gel electrophoresis or liquid chromatography to reduce complexity. We offer both bottom-up (peptide-based) and top-down (intact protein-based) MS strategies. Bottom-up proteomics is ideal for identifying hydrophobic membrane proteins, while top-down proteomics enables precise mapping of post-translational modifications.

     

    3. Quantitative Analysis of Membrane Proteins

    • We offer both label-based and label-free quantification methods. Label-based quantification methods include SILAC, TMT, and iTRAQ. For label-free quantification, we provide SRM/MRM, PRM, and SWATH-MS.

     

    4. Structural Characterization of Membrane Proteins

    • Analysis of membrane protein domains, topologies, and post-translational modifications to support functional and mechanistic studies.

     

    5. Membrane Protein–Molecule Interaction Analysis

    • Investigation of membrane protein interactions with small molecules, antibodies, ligands, or other biomolecules using techniques such as crosslinking MS, pull-down assays, or affinity capture.

     

    6. Bioinformatics Analysis of Membrane Proteins

    • We offer integrated bioinformatics analysis for membrane proteins, including data output statistics and quality control, subcellular localization and topology prediction, Gene Ontology (GO) and KEGG pathway annotation, protein-protein interaction mapping, and differential expression and enrichment analysis.
