      Technical Principles

      Our Cell Proliferation Assay Service employs the Cell Counting Kit-8 (CCK-8), which measures cell viability through mitochondrial dehydrogenase activity. In this method, the WST-8 reagent is reduced by active mitochondrial enzymes to produce an orange-colored formazan dye. The absorbance of this dye at 450 nm is directly proportional to the number of living cells, enabling quantitative, non-destructive monitoring of cell growth.

       

      By treating NIH/3T3 fibroblasts with serial dilutions of recombinant collagen and measuring absorbance across multiple time points, we generate comprehensive profiles of cell viability and growth trends. These data provide strong evidence of sample bioactivity and proliferation-promoting capacity.

       

      Analysis Workflow

      1. Sample Preparation

      Recombinant collagen is extracted into culture medium and filtered to remove particulates. Controls include a blank (ddH₂O) and a positive control (reference collagen) to ensure baseline reliability.

       

      2. Cell Culture

      NIH/3T3 mouse fibroblasts are seeded at 5×10⁴ cells/mL in a 96-well plate and cultured under standard conditions (37°C, 5% CO₂) until full adherence.

       

      3. Sample Treatment & Incubation

      Cells are treated with various concentrations of the collagen samples. After a 24-hour incubation, CCK-8 reagent is added and incubated for an additional 1–4 hours.

       

      4. Absorbance Measurement & Data Analysis

      Absorbance at 450 nm is recorded using a microplate reader. Data are analyzed to calculate relative viability and construct dose-response curves.
