Protein Identification (In-Gel, Spot, Solution) Service

In modern life science research, proteins are the core molecules that execute biological functions, widely involved in metabolic regulation, signal transduction, cellular structure, and immune response. Accurate identification of protein species and characteristics in a sample is crucial for understanding biological mechanisms, discovering biomarkers, and validating therapeutic targets. Using mass spectrometry, proteins can be digested into peptides, analyzed to generate characteristic spectra, and compared against databases for precise sequence identification. MtoZ Biolabs offers Protein Identification (In-Gel, Spot, Solution) Service that assists researchers in analyzing SDS-PAGE gel bands/spots, solution-based protein samples, or simple mixtures while ensuring cost-effective and reliable protein identification support.

    Services at MtoZ Biolabs

We utilize advanced mass spectrometry platforms, including LC-MS/MS, ESI-MS, and MALDI-TOF/TOF for protein identification across diverse sample types. Our protein identification (in-gel, spot, solution) service coverage includes:

    1. Mass spectrometry-based identification of SDS-PAGE gel bands or targeted spots.

    2. Protein identification from solution samples.

    3. Complex Mixture Protein Identification.

    4. Identification and characterization of unknown proteins in mixtures.

    5. Protein Identification from Pull-Down and Co-Immunoprecipitation Samples.

These capabilities allow us to handle a wide range of sample types, from purified single proteins to complex backgrounds.

    Analysis Workflow

    1. Sample Reception and Evaluation: Confirm sample type (gel band/spot or solution) and assess concentration, purity, and other basic parameters.

    2. Pre-treatment: Gel samples undergo destaining, reduction, alkylation, and enzymatic digestion; solution samples are directly digested.

    3. Mass Spectrometry Analysis: Acquire MS/MS spectra via LC-MS/MS or MALDI-TOF/TOF.

    4. Data Processing and Database Search: Match peptides against protein databases using professional software to generate reliable identification results.

    5. Result Review and Delivery: Conduct quality control and deliver a report including identified proteins, peptide spectra, and confidence scores.

    Applications

    1. Target Discovery and Validation: Identify differentially expressed or modified proteins in disease models to support target confirmation and mechanistic studies.

    2. Protein–Protein Interaction Studies: Identify binding partners from Co-IP or pull-down assays to build protein interaction networks.

    3. Functional Protein Analysis: Extract and identify proteins from SDS-PAGE spots for structural and functional research.

    4. Biomarker Discovery: Identify proteins in serum, tissues, or cell extracts to support preclinical biomarker discovery.

    FAQs

Q1: What is the minimum sample amount required?

A1: We recommend a total protein amount ≥ 5 μg and concentration ≥ 0.1 μg/μL. For gel samples, bands or spots should be clearly stained with relatively high purity.

Q2: Which staining method is recommended?

A2: Coomassie Brilliant Blue is preferred. Silver staining is also acceptable, though Coomassie-stained gels are more suitable for downstream identification.

Q3: How should samples be submitted?

A3: Please provide gel images along with the actual gel bands/spots in microtubes with a small amount of ddH₂O for preservation, and include sample details such as origin, concentration, and volume.

    Sample Submission Suggestions

    1. Gel Samples: Coomassie- or silver-stained bands or spots with purity ideally >90%. Submit gel photos along with samples.

    2. Solution Samples: Single proteins or lightly mixed samples with ≥ 5 μg total protein, concentration ≥ 0.1 μg/μL, and preferably >90% purity.

    3. Submission Format: Samples should be placed in clean, nuclease- and protease-free tubes with a small volume of ddH₂O. Include sample details such as concentration, volume, origin, and preparation method.

    Deliverables

    1. A standardized identification report (PDF/Word) including methods, identified proteins, peptide match information, scores, and QC notes.

    2. Raw mass spectra (optional) and peptide match tables.

    3. Data interpretation suggestions (e.g., interaction network or functional enrichment overview upon request).
