    MRM Mass Spectrometry Service

Mass spectrometry is a highly sensitive analytical technique widely used in chemistry and life sciences for the accurate measurement of the mass-to-charge ratios of molecules, enabling molecular identification and quantitative analysis. Among the many mass spectrometry techniques, multiple reaction monitoring (MRM) mass spectrometry stands out as the preferred method for targeted quantitative analysis due to its superior selectivity and sensitivity. MRM mass spectrometry enables the precise quantification of low-abundance target molecules by monitoring the reactions between specific precursor ions and product ions, making it especially suitable for analyzing complex biological samples.

MtoZ Biolabs offers professional MRM mass spectrometry service with advanced mass spectrometry instruments, such as Thermo Fisher Q Exactive and AB SCIEX TripleTOF 5600. By combining a highly skilled technical team with tailored solutions, we ensure the delivery of accurate and reliable quantitative data, facilitating breakthroughs in research, clinical studies, and drug development.

      Technical Principles

MRM mass spectrometry utilizes triple quadrupole mass spectrometry (QQQ) technology, in which three quadrupoles operate in tandem to enable precise quantification of target molecules. Following ionization of the sample in the ion source, precursor ions corresponding to the target molecules are selected in the first quadrupole (Q1), and then subjected to collision-induced dissociation (CID) in the second quadrupole (Q2) with a collision gas to generate characteristic product ions. These product ions are then filtered in the third quadrupole (Q3), and the resulting signal intensities are recorded by the detector to generate a mass spectrum. MRM’s high selectivity in ion pair screening effectively minimizes interference from non-target molecules, ensuring high reliability and a favorable signal-to-noise ratio, particularly in complex biological matrices.

      Technical Advantages

      1. MRM mass spectrometry demonstrates exceptional sensitivity, capable of detecting target molecules at the picogram level, making it ideal for trace sample analysis.

      2. The technology selectively detects precursor-product ion pairs, which effectively eliminates background interference and ensures clear, accurate results.

      3. It supports the simultaneous monitoring of multiple target molecules, enabling high-throughput, multi-sample parallel analysis, thereby increasing experimental efficiency.

      4. MRM mass spectrometry is applicable to a variety of sample types, including proteins, metabolites, and small molecules.

      Analysis Workflow

MtoZ Biolabs ensures high precision and efficiency in MRM mass spectrometry service through a comprehensive workflow supported by an advanced technical platform. This workflow includes sample preparation, targeted quantitative method development, and data analysis:

      1. Sample Preparation

Proteins are extracted from cells, tissues, or other biological samples, followed by reduction, alkylation, and enzymatic digestion to produce target peptides. Optionally, stable isotope-labeled standards can be introduced to enhance quantification accuracy and reliability.

      2. Target Selection and Method Development

Using the target protein information, specific peptide fragments are selected, and precursor-product ion transitions are optimized to develop a precise MRM detection method.

      3. Peptide Separation

High-Performance liquid chromatography (HPLC) is employed to separate enzymatically digested peptides, reducing sample complexity and providing higher resolution and sensitivity for MRM detection.

      4. MRM Mass Spectrometry Detection

Advanced triple quadrupole mass spectrometry platforms are used to selectively detect precursor and product ions, providing precise quantification of target molecules.

      5. Data Analysis and Validation

Mass spectrometry data is processed and analyzed using specialized software (e.g., Skyline). Bioinformatics tools are employed for result interpretation, generating a comprehensive analysis report.
