    Which Proteins Should You Run in WB to Test Antibody Effects on Mouse Fatty Liver

      To evaluate whether hepatic steatosis in mice is alleviated following treatment with a specific antibody, the expression levels of the following proteins associated with hepatic steatosis can be assessed by Western blot (WB):

       

      1. Proteins Related to Lipid Metabolism

      Hepatic steatosis is characterized by dysregulated lipid metabolism. Therefore, it is relevant to detect proteins involved in lipid metabolic processes, such as Fatty Acid Synthase (FAS), enzymes involved in fatty acid oxidation (e.g., CPT1, ACOX1), and fatty acid transport proteins (e.g., CD36). Alterations in the expression levels of these proteins can reflect changes in lipid metabolism.

       

      2. Inflammation-Related Proteins

      Hepatic steatosis is frequently accompanied by inflammatory responses. Proteins such as Tumor Necrosis Factor-alpha (TNF-α), Interleukin-6 (IL-6), and C-reactive protein (CRP) can be assessed to indicate the extent of inflammation.

       

      3. Oxidative Stress-Related Proteins

      Elevated oxidative stress is often observed in the context of hepatic steatosis. Relevant markers include Superoxide Dismutase (SOD), Glutathione Peroxidase (GPx), and Malondialdehyde (MDA). Changes in the expression of these proteins can provide insight into the level of oxidative stress.

       

      4. Markers of Liver Injury

      Since hepatic steatosis is often associated with liver damage, it is important to assess proteins indicative of hepatic injury, such as Alanine Aminotransferase (ALT), Aspartate Aminotransferase (AST), and Alkaline Phosphatase (ALP). Expression changes in these proteins may reflect the extent of liver injury.

       

      5. Markers of Hepatic Steatosis

      Hepatic steatosis involves excessive lipid accumulation in hepatocytes. Therefore, detection of proteins associated with lipid storage, such as Fatty Acid Binding Protein (FABP) and Triglyceride Synthase (TG Synthase), can be informative in evaluating the severity of steatosis.

       

      It is important to note that the proteins listed above represent commonly studied markers associated with hepatic steatosis. The selection of specific targets should be guided by the research objectives and experimental context. In addition, rigorous experimental design, including appropriate control groups, is essential to ensure the reliability and accuracy of the findings.
