    Membrane Proteomics Service

      Membrane proteins are specialized proteins located on the cell membrane, playing a critical role in various biological processes such as material transport, signal transduction, energy conversion, and intercellular communication. Membrane proteomics studies the structure, function, interactions, and variations of membrane proteins across diverse biological processes. The hydrophobic lipid bilayer embedding these proteins makes traditional water-soluble protein separation and purification methods ineffective, leading to challenges in extracting and purifying membrane proteins. Furthermore, their structural diversity and complexity complicate identification and characterization. However, with advances in mass spectrometry, bioinformatics, and other technologies, these challenges are gradually being addressed.

       

      MtoZ Biolabs employs Thermo Fisher's Orbitrap Exploris 240 mass spectrometer platform combined with Nano-LC chromatography to provide comprehensive subcellular proteomics analysis services, including qualitative and quantitative analysis of cellular, mitochondrial, and chloroplast membrane proteomes.

       

      Analysis Workflow

      1. Membrane Protein Isolation and Enrichment

      After cell or tissue disruption, the cell membrane or subcellular structures are isolated by centrifugation and other methods, and membrane proteins are extracted for further purification.

       

      2. Protein Digestion

      Various proteases digest membrane proteins into smaller peptide fragments for subsequent mass spectrometry analysis.

       

      3. Peptide Separation

      The digested peptides undergo further purification via HPLC.

       

      4. Mass Spectrometry Analysis

      The purified peptides are analyzed using mass spectrometry, recording mass spectrometry data like peptide ion mass-to-charge ratios.

       

      5. Mass Spectrometry Data Analysis

      The collected mass spectrometry data are cross-referenced with protein databases to identify and quantify membrane proteins.

       

      6. Bioinformatics Analysis

      Comprehensive analysis of the protein data, including protein classification, functional prediction, pathway analysis, and subcellular localization.
