DTNB (5,5'-dithiobis(2-nitrobenzoic acid), commonly referred to as Ellman's reagent) is typically dissolved in ethanol or water. During thiol group testing, the reaction of DTNB usually takes place in a near-neutral or slightly alkaline environment. This pH range is optimal because it enhances the reactivity of the thiol group with DTNB.
