How to Calculate Polysaccharide Purity?
The purity of polysaccharides is typically determined using analytical techniques such as high-performance liquid chromatography (HPLC) or gel permeation chromatography (GPC). Different analytical methods may employ distinct calculation approaches. Here, we describe a purity calculation method based on chromatographic analysis:
Sample Preparation and Analysis
The polysaccharide sample is subjected to analysis using an appropriate chromatographic system.
Peak Area Calculation
In the chromatogram, polysaccharide components and impurities appear as distinct peaks. The area of the main polysaccharide peak is determined.
Purity Calculation
Purity is typically expressed as the proportion of the main peak area to the total peak area (i.e., the sum of all detected peaks). The calculation follows the formula:

where the total peak area includes the main peak as well as all other detected peaks.
