Complex Mixture Analysis Service

In biological sample analysis, complex protein mixtures often originate from multiple sources, including cell lysates, tissue homogenates, SDS-PAGE gel bands, and immunoprecipitation (IP/Co-IP) products. Such samples place high demands on proteomic analysis, requiring not only the identification of low-abundance or rare proteins but also comprehensive coverage of all components in highly complex samples. MtoZ Biolabs offers the Complex Mixture Analysis Service that utilizes advanced multidimensional separation and high-performance mass spectrometry, enabling researchers to rapidly and accurately characterize the protein landscape of their samples while enhancing both research efficiency and data reliability.

    Services at MtoZ Biolabs

MtoZ Biolabs applies cutting-edge mass spectrometry platforms and separation technologies, integrating the following core methods:

    1. Multidimensional Protein Identification Technology (such as MUDPIT)

Complex samples are digested in solution, and peptides are separated using multidimensional chromatography strategies such as SCX and reversed-phase RPLC. High-resolution mass spectrometry is then applied to achieve broad protein coverage.

    2. GeLC-MS Method

Entire lanes from gel electrophoresis are cut into multiple segments. Proteins are separated by SDS-PAGE according to molecular weight, followed by peptide separation with RPLC, and subsequently analyzed by mass spectrometry. This approach is suitable for samples of lower complexity or when a molecular weight reference is needed.

Our complex mixture analysis service offerings include, but are not limited to:

    • Cell or tissue extracts: Study protein composition and variation in complex cell libraries or tissue samples.
    • SDS-PAGE bands: Targeted protein group analysis with molecular weight reference.
    • IP/Co-IP products: Characterize low-abundance interaction proteins enriched through immunoprecipitation.
    • Mixed sample solutions: Reveal unknown or low-concentration proteins present in mixed extracts.
    • Novel or diverse mixed samples: Characterize protein composition, abundance differences, and potential functional roles in microbial communities or co-culture systems.

    Service Advantages

    1. Multiple Mass Spectrometry Platforms

MtoZ Biolabs is equipped with a variety of mass spectrometers. Each platform has unique advantages, enabling high-resolution detection, wide dynamic range analysis, and rapid screening of complex samples. This ensures a balanced approach between proteome coverage and detection sensitivity.

    2. Multidimensional Separation Strategies

Depending on sample characteristics, MUDPIT or GeLC-MS workflows are applied to optimize separation, reduce background interference, and enhance proteome coverage, making the service suitable for different research needs.

    3. Customized Experimental Design and Flexibility

We provide a tailored complex mixture analysis service based on sample properties, research goals, and budget. Whether using solution-based MUDPIT or gel-based GeLC-MS, our services are adaptable to meet diverse project requirements.

    4. Rapid Turnaround and Detailed Reporting

We focus on efficient sample processing while delivering comprehensive and easy-to-interpret reports. Results include protein lists, sequence coverage, database search scores, and other key details that support downstream experimental design.

    FAQs

Q1: How long does it take to deliver results?

A: Typically, results are delivered within 2 to 3 weeks after sample receipt. The exact timeline depends on the sample number and complexity.

    Sample Submission Suggestions

    • Sample types: Cells, tissues (≥200 mg), liquids (≥2.5 mL), IP/Co-IP products, etc.
    • Storage and transport: Samples should be shipped on dry ice to maintain frozen conditions.
    • Information: Please provide concentration, volume, source, pretreatment details, and experimental design (such as grouping information) to facilitate analysis and interpretation.
    • Special cases: For scarce, complex, or interfering samples, please discuss with us in advance to arrange an optimized workflow.

    Deliverables

    • Experimental report: Including identified protein lists (UniProt/NCBI ID), sequence coverage, MS/MS spectrum overview, database search scores, and statistical summaries.
    • Data files: Raw mass spectrometry data (such as RAW files), spectral files, and processed data documents (such as .xlsx or .csv).
