Chloroplast Isolation and Chloroplast Protein Purification Service
Chloroplasts are essential organelles in green plants, serving as the central site for photosynthesis. In addition to their role in light-driven carbon assimilation, they possess a semi-autonomous genetic system and participate in a wide array of biosynthetic processes, including lipid production, amino acid synthesis, and terpenoid metabolism. The ability to isolate intact chloroplasts is not only a prerequisite for mechanistic studies of photosynthesis but also provides high-quality starting material for downstream applications such as chloroplast genome and transcriptome analysis, membrane system characterization, and protein–protein interaction studies.
MtoZ Biolabs offers a robust Chloroplast Isolation and Chloroplast Protein Purification Service built on optimized plant tissue workflows and efficient density gradient protocols. Our service supports a variety of research applications, including photosynthetic regulation, signal transduction, membrane proteomics, and subcellular omics.
Services at MtoZ Biolabs
MtoZ Biolabs offers specialized Chloroplast Isolation and Chloroplast Protein Purification Service designed to support a broad range of plant models and research goals. We deliver high-purity chloroplast preparations and enable targeted extraction of sub-compartments such as membranes, stroma, and thylakoids. For complex studies, we also provide customizable solutions, including multi-organelle co-isolation and tailored sample processing to support integrated multi-omics workflows.
Analysis Workflow
1. Tissue Disruption and Homogenization
Plant tissues are homogenized under optimized conditions to ensure efficient cell lysis while preserving chloroplast integrity. BSA and thiol-based stabilizers are included in the buffer to protect chloroplast membranes and minimize enzymatic degradation and oxidative damage.
2. Density Gradient Centrifugation
A three-step sucrose gradient is applied, and centrifugation parameters are finely adjusted to separate intact chloroplasts from debris and mitochondria, yielding high-purity chloroplast fractions.
3. Sub-Organelle Fractionation (Optional)
Chloroplasts are further fractionated into outer membrane, inner membrane, stroma, and thylakoid compartments using ultrasound-assisted disruption and differential centrifugation.
4. Protein Extraction and Enrichment
TCA/acetone precipitation or phenol extraction methods are used for efficient protein recovery and impurity removal. Optional membrane protein enrichment is available for specific sub-fractions.
5. Protein Quality Assessment (Optional)
SDS-PAGE, BCA assay, and Western blot can be performed to assess protein concentration, purity, and integrity to ensure compatibility with downstream applications such as proteomics or library preparation.

Applications
Built upon MtoZ Biolabs’ Chloroplast Isolation and Chloroplast Protein Purification Service, the following research areas can be effectively advanced through access to high-purity chloroplast samples and compartment-specific proteins.
FAQ
Q1: Can I submit pre-isolated chloroplasts or protein extracts for downstream analysis?
Yes. We accept multiple sample formats including fresh plant tissue, isolated chloroplasts, and protein extracts. We will provide detailed preservation and shipping guidelines to ensure sample integrity upon arrival.
Q2: Are the purified chloroplast proteins suitable for post-translational modification (PTM) analysis?
Absolutely. Our service supports enrichment and identification of PTMs such as phosphorylation, acetylation, propionylation, and succinylation via mass spectrometry. This is particularly valuable for studying regulatory mechanisms in photosynthetic efficiency and environmental response.
What Could be Included in the Report?
1. Fractionated chloroplast protein extracts (e.g., membrane, stroma, thylakoid)
2. Protein concentration and purity data (SDS-PAGE, BCA, or Western blot)
3. Standardized protocol summary and quality control documentation
4. Optional recommendations for downstream mass spectrometry-based identification
