    The principles for determining the amino acid sequence of proteins are primarily based on two main approaches: mass spectrometry and DNA/RNA sequencing.

     

    Principle of Determining the Amino Acid Sequence of Proteins by Mass Spectrometry

    1. Sample Preparation

    Proteins are enzymatically digested, typically using trypsin, to generate peptide fragments.

     

    2. Mass Spectrometry Analysis

    The resulting peptides are introduced into a mass spectrometer, where they are ionized to form charged ions. These ions are then separated and detected based on their mass-to-charge ratio (m/z).

     

    3. Data Interpretation

    The mass data obtained from the spectrometer allow for the determination of the peptide masses. Due to the high resolution of the mass spectrometer, the amino acid sequence of the protein can be inferred with high accuracy.

     

    Principle of Determining the Amino Acid Sequence of Proteins by DNA/RNA Sequencing

    1. Transcription and Reverse Transcription

    The gene encoding the protein is transcribed into RNA, which is then reverse transcribed into complementary DNA (cDNA).

     

    2. PCR Amplification

    The cDNA is amplified using polymerase chain reaction (PCR), generating multiple copies.

     

    3. Sequencing

    The PCR products are subjected to DNA sequencing to determine the nucleotide sequence of the original protein-coding gene.

     

    4. Translation

    The resulting DNA sequence is translated into the corresponding amino acid sequence of the protein.

     

    The choice between these two methods is determined by the research objectives, sample characteristics, and experimental conditions. Mass spectrometry is well-suited for already purified protein samples, whereas DNA/RNA sequencing is more appropriate when inferring protein sequences at the genetic level.
