    Overview

    S-persulfidation, also known as S-perthiolation or S-sulfhydration, is a cysteine-centered modification in which a thiol (RSH) is converted to a persulfide (RSSH). Persulfides belong to the reactive sulfur species family and are increasingly recognized as biologically relevant regulators, with both low-molecular-weight forms such as glutathione persulfide and protein-bound persulfides reported in mammalian systems.

     

    Functionally, S-persulfidation has been linked to changes in protein structure and activity and can be enriched under redox-perturbing conditions such as endoplasmic reticulum stress. Because persulfides are chemically reactive and susceptible to exchange during handling, reliable profiling typically requires controlled thiol preservation, selective persulfide capture or tag-switch strategies, and high-resolution LC-MS workflows that enable site-resolved identification and quantitative comparison across defined conditions.
