      Technical Principles

      At the core of the olink proteomics platform is PEA, a biochemical technology that combines the specificity of antibodies with the sensitivity and amplification capacity of PCR. PEA technology utilizes two proximity probes, each consisting of an antibody linked to a unique DNA oligonucleotide. When the probes bind to their target protein, they come into close proximity, allowing the DNA oligonucleotides to hybridize and form a double-stranded DNA molecule that can be extended by DNA polymerase.


      Unique sample indices are added during the subsequent PCR step, enabling efficient sample multiplexing. The resulting DNA sequences are directly quantified using real-time PCR or NGS, transforming protein presence into measurable DNA signals. With this approach, the NGS read counts are proportional to protein concentrations in the sample, allowing for accurate relative quantification.
