Analysis Workflow
1. Antibody Pair Design

2. Sample Preparation

Case Study
Plasma Proteomics Analysis of COVID-19 Patients Using Olink Assay
In a study of 306 COVID-19 patients and 78 symptomatic controls, the Olink Proximity Extension Assay (PEA) was utilized to profile 1,472 plasma proteins, revealing critical insights into disease mechanisms and progression. By leveraging the high-throughput and sensitivity of the Olink Assay, researchers identified significant upregulation of interferon (IFN) pathway proteins and chemokines such as IFN-γ, CXCL10, and CXCL11 in COVID-19 patients (Figure C–E). These proteins formed a clear disease-specific signature, distinguishing COVID-19 patients from controls and correlating with clinical severity. Through integration of proteomic data with single-cell RNA sequencing (scRNA-seq), the Olink Assay enabled precise mapping of elevated plasma proteins to immune cell origins, including T cells, monocytes, and plasmacytoid dendritic cells (Figure F). This demonstrated the assay's ability to uncover the cellular contributions driving immune dysregulation. The results showcased how Olink Assay facilitates comprehensive biomarker discovery and immune pathway analysis, providing a powerful tool for understanding disease mechanisms, identifying prognostic markers, and informing targeted therapeutic strategies for severe COVID-19.
