O6-methylguanosine is a DNA lesion caused by alkylation damage, which can be a result of both environmental factors and endogenous cellular processes. It is a methylated form of guanosine, a nucleoside consisting of guanine attached to a ribose ring. O6-methylguanosine has been recognized as a potent mutagenic lesion because it can mispair with thymine during DNA replication, leading to G:C to A:T transition mutations. It plays a key role in carcinogenesis, ageing, and other disease states. This has made its analysis an area of intense scientific and clinical interest.
The detection and quantitation of O6-methylguanosine can provide valuable information about the level of alkylating DNA damage and repair capacity in cells or tissues, as well as the potential for mutagenesis and carcinogenesis. It can also be used as a biomarker for exposure to alkylating agents, both endogenous and exogenous, and for the efficacy of anticancer therapies that target DNA alkylation damage.
