However, blood proteomics currently faces several challenges. For instance, high-abundance proteins in plasma can obscure the information from low-abundance proteins, leading to biased analysis results. Additionally, traditional mass spectrometry techniques often exhibit limited sensitivity when analyzing proteins across a wide dynamic range. Analysis Workflow
1. Sample Collection and Processing
Blood samples are collected using EDTA anticoagulant tubes, gently mixed, and centrifuged at 4°C for 10 minutes to separate the plasma.
2. Protein Extraction and Purification
The upper plasma layer is transferred to a centrifuge tube, protease inhibitors are added, and the sample is rapidly frozen in liquid nitrogen to maintain quality.
3. Enzymatic Digestion and Peptide Generation
The extracted proteins are digested to produce peptides suitable for mass spectrometric analysis.
4. Mass Spectrometry Analysis
High-throughput mass spectrometry devices conduct LC-MS/MS analysis to comprehensively capture protein data.
5. Data Processing and Analysis
Advanced software, along with bioinformatics techniques, is employed for precise protein identification and quantification.
