      Case Study

      Colorectal cancer (CRC) is highly heterogeneous, and despite extensive genomic studies, clinically actionable biomarkers remain limited. A proteogenomic study systematically analyzed paired tumor and normal tissues, integrating phosphoproteomics and genomics to identify cancer-associated proteins, phosphosites, and therapeutic targets. Phosphoproteomics linked Rb phosphorylation to increased proliferation and reduced apoptosis, suggesting a rationale for targeting Rb phosphorylation in CRC treatment. Additionally, MSI-H tumors exhibited increased glycolysis and reduced CD8+ T cell infiltration, highlighting glycolysis as a potential target to enhance immune checkpoint therapy. The study also identified novel cancer-specific phosphosites, advancing biomarker discovery and precision oncology.
