The resulting derivatives are subjected to gradient elution using HPLC. This process separates different amino acid derivatives, and their concentrations are quantified using external standards.     Sample Submission Suggestions

    1. Sample Types

    Samples with excessively high salt concentrations should be avoided.

     

    3. Buffer Solutions

    Avoid buffers containing large amounts of primary amines, such as TRIS or glycine, as these can interfere with ninhydrin reactions and result in inaccurate quantification of specific amino acids.

     

    4. Impurities

    Ensure that samples are free from impurities such as fingerprints or dust, as these contaminants may contain human proteins that can interfere with detection results.

     

    5. Sample Amount

    The recommended sample amount is >2 mg. Please provide sufficient solid material to allow accurate weighing during the analysis. You may choose one or three replicates for amino acid determination. For highly accurate protein concentration measurements, we recommend triplicate biological replicates.

    1. Simultaneous Analysis of 26 Physiological Amino Acids and Total Cysteine/Homocysteine in Plasma Using HPLC 

    The quantification of plasma amino acid levels has significant clinical implications for disease diagnosis, therapeutic monitoring, nutritional assessment, and physiological research. The study employed HPLC with 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate (AQC) as a pre-column derivatization reagent to simultaneously analyze 26 physiological amino acids in plasma. Amino acid HPLC analysis service was applied in metabolic laboratories to establish reference ranges for plasma amino acids and identify metabolic disorders such as classical homocystinuria, cobalamin deficiency, nonketotic hyperglycinemia, hyperprolinemia, ketotic hyperglycinemia, urea cycle defects, and maple syrup urine disease. HPLC not only provided quantitative data on amino acids in plasma but also expanded its application to identify low-level amino acid metabolites, demonstrating significant potential for clinical research.
