      Acylation dynamically regulates cellular metabolism, gene expression, and stress responses. Common types include: acetylation, palmitoylation, malonylation, succinylation, glutarylation, propionylation, crotonylation, and β-hydroxybutyrylation. Dysregulated acylation contributes to cancer, neurodegenerative disorders, cardiovascular diseases, and infections. For example, hyperacetylation of histones/nonhistone proteins drives oncogenic signaling, while defective palmitoylation disrupts neuronal synaptic function. Targeting acyltransferases (e.g., p300/CBP), deacylases (e.g., sirtuins, HDACs), or acylation "reader" proteins offers therapeutic potential. Small-molecule inhibitors (e.g., HDACi in cancer) and metabolites (e.g., NAD⁺ boosters for sirtuin activation) are under clinical development.
