7α,25-Dihydroxycholesterol (7α,25-OHC) is a dihydroxylated derivative of cholesterol, characterized by the presence of two hydroxyl (–OH) groups at the 7th and 25th carbon positions of the cholesterol molecule. In biological systems, 7α,25-OHC acts as a unique signaling molecule with significant biological activity, particularly within the immune system. It is an endogenous high-affinity ligand for the G protein-coupled receptor EBI2 (also known as GPR183). EBI2 is highly expressed in various immune cells, such as B cells and dendritic cells, and plays a critical role in regulating the migration, distribution, and function of these immune cells through its interaction with 7α,25-OHC. The critical role of 7α,25-OHC and its receptor EBI2 in modulating immune cell function makes it a potential target for drug development. Quantitative analysis of 7α,25-OHC can help evaluate its level changes under different pathological conditions, assisting in the validation of EBI2-related drug targets and providing a basis for designing new immunomodulatory drugs.
The biosynthesis of 7α,25-OHC involves multiple enzymatic steps, including those catalyzed by CYP7A1 and CYP27A1. Quantitative analysis of 7α,25-OHC and its metabolites can help reveal the activity of these metabolic pathways in different tissues, explore their changes under physiological and pathological conditions, and provide insights for studying the mechanisms underlying metabolism-related diseases.
