5-methyluridine (m5U) is a pyrimidine nucleoside, featuring a methyl group attached to the 5-position of the uracil base. It is one of the most common modifications found in cellular RNA. m5U is almost universally present at position 54 in eukaryotic and bacterial tRNA, where it plays a crucial role in stabilizing the structure of these molecules. The same “T loop” motif is also observed in various other forms of non-coding RNA, including tmRNA and rRNA.
Research has shown that the absence of tRNA modifications like m5U typically does not lead to drastically different phenotypes. However, understanding the nuances of 5-methyluridine and its modifications is essential for deciphering RNA biology and its impact on cellular processes, such as translation and stress responses. Given its significance, m5U is emerging as a potential biomarker for various diseases, including cancer.
