5-Methylcytosine (5mC) is an important biomolecule and a form of DNA modification, with the chemical formula C₄H₅N₃O. It is formed through the DNA methylation of cytosine, typically occurring at the cytosine residues of CpG dinucleotides (CpG dinucleotide refers to a DNA sequence composed of one cytosine (C) and one guanine (G), with a phosphodiester bond connecting the C and G). 5-Methylcytosine plays a crucial role in regulating gene expression, cell differentiation, and development. It is often associated with transcriptional repression by recruiting repressive factors or altering chromatin conformation to inhibit gene expression. Analyzing the levels of 5-methylcytosine can reveal its role in gene silencing and transcriptional regulation, aiding in the understanding of the complex mechanisms of gene expression. Additionally, examining the levels of 5-methylcytosine and its metabolites supports the implementation of personalized medicine by providing biomarkers for therapeutic intervention.
