5-hydroxymethylcytidine (hm5C) is a derivative of cytidine, found in the DNA of cells. It is an offspring of 5-methylcytidine (5mC), which is a product of an enzymatically controlled process of DNA methylation. The occurrence of hm5C is a significant epigenetic modification that is pervasively present in human tissues, especially abundant in the brain and embryonic stem cells. hm5C plays a critical role in a myriad of biological processes, such as gene expression, DNA methylation dynamics, and embryonic development. Moreover, abnormal hm5C levels have been associated with various pathologies, including cancer, neurological disorders, and autoimmune diseases.

The understanding of how hm5C fits into the epigenetic landscape is still in its infancy. It's essential to develop an accurate, sensitive, and high-throughput method for the quantitative analysis of hm5C to garner a better understanding of its biological function and significance in health and disease.
At MtoZ Biolabs, we utilize LC-MS/MS for the analysis of hm5C, offering quantification for hm5C and its derivatives. Our well-established method allows us to detect hm5C in a broad range of biological samples, providing accurate, reliable, and rapid results.
