4D Glycosylation Proteomics Service

MtoZ Biolabs offers 4D Glycosylation Proteomics Service to provide detailed and high-resolution insights into the glycosylation of proteins across complex biological samples. Using advanced ion mobility mass spectrometry, this service enhances the detection and characterization of glycosylated peptides, even at low abundance. By adding ion mobility as a fourth dimension to 3D proteomics, we improve the sensitivity and specificity of glycosylation site identification, offering a powerful tool for glycoproteomics studies, biomarker discovery, and functional glycoscience.

    What is 4D Glycosylation Proteomics?

Protein glycosylation is an enzymatically driven modification in which glycans are covalently attached to specific amino acid residues, influencing protein folding, trafficking, stability, and molecular recognition. In proteomics workflows, glycosylation is challenging because glycan structures are highly heterogeneous, glycopeptides often occur at low abundance, and glycoforms can be difficult to distinguish in crowded spectra.

4D proteomics adds ion mobility to the conventional 3D dimensions used in LC-MS-based proteomics (RT, m/z, and intensity). Ion mobility separates ions by shape and collision cross-section, providing an additional layer of selectivity that helps reduce co-elution interference and improves discrimination of closely related species. For glycoproteomics, this translates into more confident glycopeptide detection and improved coverage, particularly when glycoforms or near-isobaric species compete within the same chromatographic window.

Projects typically use 4D glycosylation proteomics to

    • Map glycosylation sites and profile glycoform distributions across conditions
    • Characterize N-glycosylation and O-glycosylation changes linked to phenotype or treatment
    • Improve discovery in low-abundance or high-dynamic-range samples
    • Support mechanism studies involving glycosylation-dependent signaling, immunity, or cell-cell interactions
    • Generate candidate targets for downstream verification, including targeted MS follow-up
