    10x TCR Sequencing Service

10x TCR Sequencing Service is an advanced high-throughput approach for characterizing T-cell receptor (TCR) repertoires at the single-cell level. T-cell receptors are essential components of the adaptive immune system, enabling T cells to recognize and respond to a vast array of antigens. These receptors, located on the surface of T cells, mediate antigen recognition by interacting with peptide-major histocompatibility complex (pMHC) molecules, initiating downstream immune responses.

A defining feature of TCRs is their immense diversity, known as the TCR repertoire diversity, which arises from V(D)J recombination and somatic hypermutation. This diversity ensures that the immune system can recognize virtually any foreign or mutated antigen, forming the basis of adaptive immunity. Characterizing the TCR repertoire provides invaluable insights into immune responses in infections, autoimmune disorders, and cancer immunotherapy. The 10x TCR Sequencing Service offers a powerful, high-resolution approach to unraveling TCR diversity at the single-cell level, facilitating breakthroughs in immunology and translational medicine.

      Case Study

Unraveling Heterogeneous Antitumor Immunity with 10x TCR Sequencing Service

Antitumor immune responses vary significantly among individuals, influencing therapeutic outcomes in cancers such as head and neck squamous cell carcinoma (HNSCC). To investigate the underlying mechanisms, researchers developed a unique HNSCC mouse model and employed the 10x TCR Sequencing Service to analyze CD8 tumor-infiltrating lymphocytes (TILs). Despite genetic uniformity, ~25% of mice eradicated tumors without intervention, driven by CD8 T-cell activity. Single-cell TCR sequencing revealed distinct TCR clonotypes in regressors and progressors, with minimal overlap. Top clonotypes clustered in different activation states, highlighting the influence of the tumor microenvironment on T-cell responses. These findings underscore the highly individualized nature of antitumor immunity, emphasizing the role of 10x TCR Sequencing Service in decoding TCR diversity. This approach provides critical insights for developing personalized cancer immunotherapies.

      FAQ

Q: What are the differences between TCR and BCR?

TCR (T-cell receptor) and BCR (B-cell receptor) are critical components of adaptive immunity with distinct roles. TCR is expressed on T cells and recognizes processed antigens presented by MHC molecules, requiring cell-cell interaction. BCR is expressed on B cells and directly binds unprocessed, native antigens in soluble or membrane-bound forms. Structurally, TCR comprises α and β (or γ and δ) chains, while BCR is an immunoglobulin with heavy and light chains. Functionally, TCR activation mediates cellular immune responses, whereas BCR activation leads to antibody production, bridging humoral immunity. Both are essential for immune system precision and diversity.
