Targeted Proteomics
Targeted proteomics is a mass spectrometry method that focuses on studying a specific group of proteins or peptides, which is different from traditional global proteomics analysis. Targeted proteomics usually employs selective reaction monitoring (SRM) or multiple reaction monitoring (MRM) mass spectrometry techniques to specifically detect and quantify specific peptides.
Advantages
1. High Sensitivity and Specificity
Since it only focuses on a predefined group of peptides, the detection sensitivity and specificity are very high.
2. Accurate Quantification
Accurate quantification can be achieved using appropriate internal standards and calibration curves.
3. Rapid Analysis Time
Since it only focuses on a small group of targets, the analysis time is relatively short.
Limitations
1. Limited Target Quantity
Typically, only a small number of target peptides can be monitored.
2. Prior Knowledge Is Required
It is necessary to know in advance the target proteins or peptides to be studied.
Targeted proteomics provides researchers with an efficient and accurate method to study and quantify specific target proteins or peptides. Due to its high sensitivity and specificity, targeted proteomics is widely used in biomarker validation, drug metabolism research, functional proteomics and many other fields. Compared with global proteomics, it is more focused, but its scope is relatively narrow.
