    PCR Based Antibody Sequencing Service

Antibodies are crucial molecules employed by the immune system to recognize and neutralize foreign substances. Antibody sequencing is a fundamental technique used to determine the amino acid sequence of antibodies, thus elucidating their structure and function. This technology has wide-ranging applications, including drug development, disease research, and personalized medicine.

      Services at MtoZ Biolabs

MtoZ Biolabs has developed an advanced PCR-based method for antibody sequencing. This technique employs specifically designed degenerate primers to amplify antibody genes from hybridoma cells, ensuring the capture of critical nucleotide sequences. This process facilitates the stable expression and production of target antibodies in subsequent stages. Our methodology includes detailed degenerate primer construction and precise sequencing strategies to deliver rapid and accurate sequencing of both antibody variable regions and full-length genes. MtoZ Biolabs provides comprehensive antibody sequencing services, from initial analysis to final reporting, tailored to meet the specific needs of each client.

Figure 1. The Workflow of PCR Based Method for Antibody Sequencing

      Analysis Workflow

The PCR-based antibody sequencing method employs polymerase chain reaction (PCR) technology to amplify antibody gene fragments using specific primers. The amplification products are then sequenced using Sanger sequencing or next-generation sequencing (NGS) technologies. The workflow includes the following steps:

      1. Sample Extraction

Extracting RNA or DNA from sources such as cells, tissues, or blood samples.

      2. Reverse Transcription and PCR Amplification

For RNA samples, cDNA is synthesized via reverse transcription, followed by PCR amplification using specific primers. Then direct PCR amplification is performed for DNA samples.

      3. Purification of Amplified Products

Purifying PCR products using gel electrophoresis or other purification techniques to eliminate non-specific products.

      4. Sequencing

Sequencing the purified PCR products using Sanger or NGS technologies to obtain the nucleotide sequence of the antibody gene.

      5. Data Analysis

Analyzing sequencing data to determine the amino acid sequence of the antibody.
