      Case Study

      High-Specificity Detection of E. coli in Fish Samples Using MALDI Microbial Identification Service

      In this study, E. coli served as a model bacterium to evaluate the effectiveness of a novel protocol combining anti-E. coli magnetic beads with MALDI-TOF analysis. Magnetic beads successfully captured E. coli from fish sarcoplasmic protein (FSP) and muscle protein extracts with high reproducibility and sensitivity. A partial least-squares discriminant analysis (PLS-DA) plot showed clear discrimination of E. coli in matrices of varying complexity, including water, fish muscle surface, and protein extracts. The method achieved excellent accuracy (RSD = 3%) and identified 10 biomarker ions specific to E. coli, even in the presence of interfering bacteria. This study highlights the potential of the MALDI Microbial Identification Service as a reliable tool for enhancing food safety and ensuring the accurate detection of microbial contaminants in complex food matrices.
