When differentially labeled peptides enter the mass spectrometer, they produce distinct peak clusters with specific mass shifts in the spectrum. By comparing the intensity of these peaks, researchers can achieve a deep quantitative analysis of relative protein levels, accurately measuring subtle differences in protein expression between samples. Moreover, when internal standards or known concentration reference standards are included, the captured mass spectrometry signals can be used to precisely determine absolute protein quantities, enabling absolute quantification.
