      Exosomal mRNA Microarray Reveals RNA-Mediated Communication Between Mast Cells and Progenitor Cells

      A pioneering study investigated whether exosomes derived from human mast cells could mediate intercellular communication through RNA transfer. Using exosomes secreted by the HMC-1 human mast cell line, researchers employed both mRNA and microRNA microarray platforms to profile their RNA content. The analysis revealed approximately 1,800 distinct mRNAs and 116 microRNAs within the exosomal cargo, representing about 15% of the mRNA population in the donor cells. Functional assays, including co-culture, confocal microscopy, and radioactive labeling, confirmed that these RNA-containing exosomes were not only taken up by neighboring mast cells but also delivered their RNA payloads to CD34+ hematopoietic progenitor cells. This demonstrated that exosomal shuttle RNA (esRNA) serves as an active mediator of cell-to-cell communication, potentially influencing cell differentiation and maturation processes.
