    10x BCR Sequencing Service

The 10x BCR Sequencing Service utilizes cutting-edge technology to enable high-resolution analysis of B-cell receptor (BCR) diversity. B-cell receptors play a crucial role in the immune system by enabling B cells to recognize and respond to antigens. Each B cell expresses a unique BCR, contributing to the vast diversity of antibodies that can be generated to combat pathogens. This diversity is essential for adaptive immunity, making BCR sequencing a valuable tool for understanding immune responses and antibody evolution.

The 10x BCR Sequencing Service leverages the advanced capabilities of the 10x Genomics platform, a cutting-edge tool that provides high-throughput, precise, and comprehensive sequencing of BCRs, enabling a deeper understanding of immune diversity and antibody dynamics. Using the 10x Genomics technology, it is possible to sequence entire BCR repertoires with single-cell resolution, allowing for the identification of both heavy and light chain sequences in their full diversity.

The principle behind the 10x BCR sequencing involves single-cell RNA sequencing (scRNA-seq) integrated with unique molecular identifiers (UMIs) that help to capture and resolve rare and diverse BCR sequences from complex immune samples. The process starts with isolating single B cells, followed by the amplification and sequencing of their respective BCR genes. The 10x Genomics platform's ability to analyze thousands of single B cells in parallel ensures that researchers can obtain comprehensive data on BCR diversity and clonal relationships.

      Case Study

Leveraging 10x BCR Sequencing Service to Investigate B-Cell Repertoire Dynamics in Severe COVID-19

A multi-omics study utilized the 10x BCR sequencing technology to investigate B-cell responses in severe COVID-19. The study aimed to explore BCR repertoire dynamics and identify potential biomarkers for disease severity. Over 358,000 cells from 14 COVID-19 patients were analyzed at multiple time points using both bulk BCR sequencing and single-cell BCR sequencing (scBCR-seq). The results revealed an increase in B-cell clonality early in infection, which normalized during recovery. A significant expansion of IgA and IgG plasmablasts was observed, particularly in the early stages of disease. Additionally, analysis of IGHV gene usage showed a skewed representation of IGHV3-30 and IGHV3-23, indicating immune system adaptation to SARS-CoV-2. This study highlights the power of the 10x BCR Sequencing Service in providing critical insights into immune responses, offering a foundation for developing biomarkers and targeted treatments for COVID-19.

      FAQ

Q: What is the role of UMI (Unique Molecular Identifiers) in the 10x BCR Sequencing Service?

Unique Molecular Identifiers (UMIs) are used to tag individual cDNA molecules, ensuring accurate quantification of each transcript. UMIs help distinguish between true biological variation and PCR-induced errors, significantly reducing amplification biases. By applying UMIs, the 10x BCR Sequencing Service enhances sequencing precision, particularly in complex immune repertoires, allowing for reliable detection of rare B-cell clones and improving overall data quality in BCR repertoire analysis.
