What Is TMT Isobaric Labeling?

    TMT (Tandem Mass Tag) isobaric labeling is a mass spectrometry–based technique widely employed in proteomics research for the quantitative assessment of relative protein abundance. This approach utilizes specialized isotopic tags that possess identical overall masses but generate distinct reporter ions upon fragmentation. In this way, protein abundance across multiple samples can be simultaneously compared.

    The TMT isobaric labeling workflow consists of the following steps:

    1. Protein Extraction

    Proteins are isolated from various biological samples.

    2. Protein Digestion

    The extracted proteins are digested into peptides using proteolytic enzymes, such as trypsin.

    3. Labeling

    Peptides from each sample are labeled with specific TMT reagents. Although the reagents share identical masses, they yield unique reporter ions upon fragmentation.

    4. Mixing

    The labeled peptide samples are pooled together.

    5. Mass Spectrometry Analysis

    The combined peptide mixture is analyzed by liquid chromatography–tandem mass spectrometry (LC–MS/MS). The mass spectrometer detects peptide masses and their fragment ions, including the reporter ions.

    6. Data Analysis

    Quantitative information on relative protein abundance across samples is obtained by comparing the signal intensities of the reporter ions.

    TMT isobaric labeling offers several advantages, including high sensitivity, high throughput, and the capability of multiplexed sample analysis. Nonetheless, certain limitations exist, such as interference from isotopic impurities and the requirement for complex computational analysis. Despite these challenges, TMT isobaric labeling remains a critical and powerful tool for quantitative proteomics.
