    Yeast 2 Hybrid Analysis Service

In Y2H, these domains are functionally split and fused to two proteins of interest—referred to as the "bait" (fused to the DBD) and the "prey" (fused to the AD). When the bait and prey proteins physically interact within the yeast nucleus, the DBD and AD are brought into proximity, reconstituting a functional transcription factor. This reconstitution activates the expression of a reporter gene, such as GFP or lacZ, which can be detected through selective growth media or colorimetric assays. The strength and specificity of the interaction can also be quantitatively assessed by measuring reporter gene activity.
