Selected reaction monitoring mass spectrometry is an analytical technique based on tandem mass spectrometry, offering high sensitivity and precise quantification. It is widely utilized in proteomics, metabolomics, pharmacokinetics, and environmental science. By leveraging the high selectivity of tandem mass spectrometry, SRM enables highly specific detection through precise selection of precursor ions and their characteristic fragment ions. Selected reaction monitoring mass spectrometry primarily relies on triple quadrupole mass spectrometers. The technique's core strength lies in the ability of the triple quadrupole system to isolate and analyze specific ion pairs: the first quadrupole (Q1) selects the precursor ion, the second quadrupole (Q2) fragments it via collision-induced dissociation (CID), and the third quadrupole (Q3) detects specific fragment ions. This dual mass filtering process ensures exceptional quantitative precision, making SRM particularly effective for targeted protein and metabolite analysis.
Despite its advantages in specificity and accuracy, selected reaction monitoring mass spectrometry presents certain challenges. First, optimizing precursor-product ion pairs and collision energy for each target molecule requires extensive method development, limiting its applicability to targeted analysis rather than unknown molecule screening. Additionally, because SRM relies on predefined ion pairs, its throughput is constrained, making simultaneous detection of a large number of molecules difficult.
